Pharmacokinetics

Bioavailability Peak plasma level Plasma half-life Active metabolites Elimination
variable* 60-95%* 2 to 6 hours 10 hours (2 - 24 hours) moderately important predominantly hepatic
*There are probably greater discrepancies of bioavailability between the different manufacturers' preparations. On average 3.5 mg of a micronised preparation are said to be equal to 5 mg of a non-micronised preparation. The duration of the effect is 18 to 24 hours.

*Micronised preparations: initially 0.875 to 3.5 mg/day; later an increase to 14 mg/day is possible. When switching from one preparation to another it is recommended to re-evaluate the dose based on the blood glucose levels. Increase dose only gradually!
**Take 15 to 30 minutes before breakfast. Daily doses of more than 10 mg should be divided in two.
