Thioridazine: Pharmacology

Thioridazine is a phenothiazine. The neuroleptic effect of these drugs is probably based on an antagonism at the dopamine (D2) receptors in the mesolimbic system. An analogous effect in the nigrostriatal system is probably responsible for the extrapyramidal side-effects. Thioridazine has pronounced anticholinergic activity. It also stimulates prolactin secretion. Other effects (e.g. at the alpha1-adrenoreceptors) hardly have clinical relevance.
